沙棘果油及沙棘籽油对大鼠放射性皮肤损伤的影响
【摘要】 目的 观察沙棘果油及沙棘籽油对大鼠放射性皮肤损伤的影响。方法 实验用深部X线治疗机,每次以3000cGy的剂量,照射两次,造成大白鼠臀部放射性皮肤损伤。待动物症状明显后,将Ⅱ度皮肤损伤的动物随机分为6组,即阴性对照组、沙棘果油高剂量组、沙棘果油低剂量组、沙棘籽油高剂量组、沙棘籽油低剂量组以及阳性对照组。沙棘果油及籽油高剂量组,每天在损伤部位涂药两次,而其低剂量组每天涂药一次,阴性对照组每天涂灭菌的注射用水一次,阳性对照组每天涂SolcoserylJelly软膏一次。结果 肉眼观察结果显示,连续用药10d后,沙棘果油、籽油及SolcoserylJelly组动物的局部红斑、水肿明显减轻,创面分泌物明显减少,用药2周后,上述症状明显消退,与阴性对照组比较,明显好转;另外,沙棘果油、籽油及SolcoserylJelly组的创面愈合时间明显缩短。光学显微镜观察结果显示,沙棘果油、籽油及SolcoserylJelly组的皮肤损伤明显减轻,炎症反应亦明显减轻,创面修复明显。电子显微镜观察结果显示,阴性对照组的超微结构明显受损,而沙棘果油、籽油及SolcoserylJelly组的损伤明显减轻。结论 沙棘果油及沙棘籽油对大鼠放射性皮肤损伤有明显的治疗作用。

ABSTRACT: Objective To observe the effects of sea buckthorn pulp oil and sea buckthorn seed oil on radiation skin lesions of rats. Methods The experiment used deep xray therapeutic machine to irradiate the rats twice, and the irradiation dosage was 3000cGy each time. This caused radiation skin lesions to rats buttock. After the symptoms became obvious, the rats suffering from Grade Ⅱ skin lesions were randomly divided into six groups: negative control group, highdose sea buckthorn pulp oil group, lowdose sea buckthorn pulp oil group, highdose sea buckthorn seed oil group, lowdose sea buckthorn seed oil group, and positive control group. The oil was applied to the wounded area twice a day for highdose pulp oil group and highdose seed oil group, and it was applied once a day for the lowdose groups. For the negative control group, sterilized injection water was applied once a day, and SolcoserylJelly ointment was applied once a day for the positive control group. Results The results from naked eye observation showed that after medication for 10 consecutive days, the local erythema and edema of the groups using sea buckthorn pulp oil and seed oil and SolcoserylJelly ointment abated, and the secretions of the wounded surface reduced considerably. After medication for two weeks, obvious disappearance of the above symptoms was observed; compared with the negative control group, the condition improved considerably. In addition, the healing time of the wounded area of the rats was markedly shortened for the pulp oil groups, seed oil groups, and the SolcoserylJelly group.Optical microscopic observation showed that the injuries in the pulp oil group,seed oil group,and the SolcoserylJelly group were significantly reduced, inflammatory reaction was obviously reduced, and the restoration of the wounded areas was obvious. Electron microscopic observation showed that the ultrastructure of the negative group was obviously damaged, but that of the pulp oil groups, seed oil groups, and SolcoserylJelly group reduced significantly. Conclusion The above results suggest that sea buckthorn pulp oil and sea buckthorn seed oil have a marked therapeutic effect on radiation skin lesions in rats.

沙棘籽油及沙棘果油是从胡颓子科植物沙棘的籽及果肉中提取。据报道,沙棘油可用于治疗溃疡病、烧、烫伤等多种疾病[1]。张文禄等[2]报道沙棘油对肿瘤患者放射治疗所引起的急性放射性皮炎、黏膜炎症及非放射性的皮肤炎症、溃疡、伤口愈合不良等都有较好的疗效。高力英等[3]报道沙棘油对患者放射性皮肤损伤有防治作用。本实验观察了用超临界萃取法提取的沙棘籽油及沙棘果油对大鼠放射性皮肤损伤的影响。

1.1 实验动物
健康的SD大鼠,雌性,体重(200±20)g,由西安交通大学医学院实验动物中心提供。
1.2 药品
沙棘籽油(sea buckthorn seed oil, SBSO)及沙棘果油(sea buckthorn pulp oil, SBPO),用超临界萃取法提取,由芬兰土尔库大学提供。SolcoserylJelly软膏,由瑞士素高大药厂生产。
